Highly Sensitive and Specific Treponema pallidum Antibody Test - Alternative Method in Screening for and Diagnosis of Syphilis Infection
Principle of the Test:
Wantai anti-TP ELISA uses polystyrene microwell strips pre-coated with recombinant Treponema pallidum antigens expressed in E. coli. The sample is incubated in the microwells together with recombinant TP antigens conjugated to horseradish peroxidase (HRP). In case of presence of anti-TP in the sample, the pre-coated and conjugated antigens will be bound to the two variable domains of the antibody and during incubation, the specific antigen-antibody immunocomplex is captured on the solid phase. After washing to remove unbound sample proteins and conjugates, Chromogen solutions (TMB) are added to the wells. In presence of the antigen-antibody-antigen(HRP) "sandwich" complex, the colorless Chromogens are hydrolyzed by the bound HRP conjugate to a colored product that can be measured and is proportional to the amount of antibody in the sample.
